Rational Distribution of Water Resources of North China and South-to-North Water Diversion

Abstract:

According to the present status of China´ s water resources and the needs of sustainable development, it is imperative and possible to carry out the south-to-north water diversion project, that is to divert water from the Yantze River valley, which has plentiful water resources, upward to the north part of China.

The principle of south-to-north water diversion should be as follows: Establish the sociaty of water-saving, do not supply water according to the “exaggerated needs”; distribute water rationally, and pay attention to tapping the potential; the east and weet water diversion routes are existing side by side and playing a part together, not irreconcilable to each other; make budget estimate in a practical and realistic way, avoiding endless supplementary budgets; the partial and local interests must be subordinated to the interests of the whole, and the temporary interests, subordinated to the long-term enes; implement unified management, scientific allocation and the charginy system for use of water resources; to divert water from the southwest region is not realistic, better to put off.

Rational design on photoelectrodes and devices to boost photoelectrochemical performance of solar-driven

Abstract: As an eco-friendly, efficient, and low-cost technique, photoelectrochemical water splitting has attracted growing interest in the production of clean and sustainable hydrogen by the conversion of abundant solar energy. In the photoelectrochemical system, the photoelectrode plays a vital role in absorbing the energy of sunlight to trigger the water splitting process and the overall efficiency depends largely on the integration and design of photoelectrochemical devices. In recent years, the optimization of photoelectrodes and photoelectrochemical devices to achieve highly efficient hydrogen production has been extensively investigated. In this paper, a concise review of recent advances in the modification of nanostructured photoelectrodes and the design of photoelectrochemical devices is presented. Meanwhile, the general principles of structural and morphological factors in altering the photoelectrochemical performance of photoelectrodes are discussed. Furthermore, the performance indicators and first principles to describe the behaviors of charge carriers are analyzed, which will be of profound guiding significance to increasing the overall efficiency of the photoelectrochemical water splitting system. Finally, current challenges and prospects for an in-depth understanding of reaction mechanisms using advanced characterization technologies and potential strategies for developing novel photoelectrodes and advanced photoelectrochemical water splitting devices are demonstrated.

Rational Understanding about Renaissance Path of China's Road and Bridge Engineering

Abstract: At the turn of this century, China began the largest road and bridge development in the world, which has virtuously interacted with the social and economic development and has elevated the engineering technology to world-class level. Surveying the development of world transportation, and comparing the road and bridge construction efforts of Europe, U.S.A, Japan and China, general experiences of success can be derived as follows: It is greatly beneficial to give priority to transportation for economic development; it takes national strategy and social action to build up a transportation network; demand and innovation are the driving forces to upgrade road and bridge technology; management innovation and quality assurance can greatly improve development caliber. While rationally acknowledging China's transportation infrastructure achievements, we should reflect on issues with Chinese characteristics that have affected quality and efficiency, expose all construction practices that violate scientific law, overall management defects in road and bridge construction and maintenance, and search for new ways to strengthen social management and the creation of a healthy soft environment.

Rational Design of and Mechanism Insight into an Efficient Antifreeze Peptide for Cryopreservation

Abstract: The development of effective antifreeze peptides to control ice growth has attracted a significant amount of attention yet still remains a great challenge. Here, we propose a novel design method based on an in-depth investigation of repetitive motifs in various ice-binding proteins (IBPs) with an evolution analysis. In this way, several peptides with notable antifreeze activity were developed. In particular, a designed antifreeze peptide named AVD exhibits ideal ice recrystallization inhibition (IRI), solubility, and biocompatibility, making it suitable for use as a cryoprotective agent (CPA). A mutation analysis and molecular dynamics (MD) simulations indicated that the Thr6 and Asn8 residues of the AVD peptide are fundamental to its ice-binding capacity, while the Ser18 residue can synergistically enhance their interaction with ice, revealing the antifreeze mechanism of AVD. Furthermore, to demonstrate the cryoprotection potential of AVD, the peptide was successfully employed for the cryopreservation of various cells, which demonstrated significant post-freezing cell recovery. This work opens up a new avenue for designing antifreeze materials and provides peptide-based functional modules for synthetic biology.

Abstract:

At the turn of this century,China began to undertake the largest scale road and bridge construction in the world,which has virtuously interacted with the social and economic development and has brought the construction technology up to the world advanced level. Looking back to the world transportation development and comparing the road and bridge construction in Europe,USA,Japan and China,the common successful experiences can be obtained as follows: it has become the value orientation for transportation engineering to give the priority to the transportation for economic development;it has become the national strategy and social action to build up great transportation artery;demand and innovation are the driving force to upgrade road and bridge construction technology;management innovation and quality control have upgraded the project construction quality. While rationally recognizing the achievements in the transportation construction of our country,we should review the deep rooted problems with China’s characteristics that have affected construction quality and efficiency;reveal any construction behavior that violates scientific law and extensive management in road and bridge construction and maintenance;probe into the new challenges to strengthen social management and create healthy soft environment.

An Intelligent Manufacturing Platform of Polymers: Polymeric Material Genome Engineering Perspective

Abstract:

Polymeric materials with excellent performance are the foundation for developing high-level technology and advanced manufacturing. Polymeric material genome engineering (PMGE) is becoming a vital platform for the intelligent manufacturing of polymeric materials. However, the development of PMGE is still in its infancy, and many issues remain to be addressed. In this perspective, we elaborate on the PMGE concepts, summarize the state-of-the-art research and achievements, and highlight the challenges and prospects in this field. In particular, we focus on property estimation approaches, including property proxy prediction and machine learning prediction of polymer properties. The potential engineering applications of PMGE are discussed, including the fields of advanced composites, polymeric materials for communications, and integrated circuits.

Study on Mechanical Behavior of a Prestressed ConcreteCable-stayed Bridge in Construction State

Abstract:

Because of the increasing span of concrete cable-stayed bridge,  the construction simulation recently becomes more focusing on both the design and construction.  This paper introduces the optimization theory and method into the calculation of cantilever construction of girders. The example bridge is a long-span prestressed concrete cable-stayed bridge with a 370 m central span length under construction in China. Using spatial nonlinear FEM,  the actual cantilever construction process of concrete box-shaped girders is simulated,  the tensioning force of cables as well as the prearranged height of girders in construction state is obtained. At the same time, the influence of structural geometric nonlinear characteristics and physics nonlinear characteristics in the cable force optimum are investigated,  the measures to reduce the influence are put forward.

Rational Design, Over-expression and Characterization of a Novel Staphylokinase with Lower Tendency of

Abstract:

Staphylokinase has strong tendency to polymerize, which may hamper its clinical use. On the basis of crystal structure of staphylokinase, the dimer structure was modeled by software GRAMM V1.03 with high-resolution generic docking parameters. Two kinds of binding interfaces were generated, mainly linked by hydrophobic interaction and hydrogen bonds. The computer model of staphylokinase dimer provides a template for the design of improved staphylokinase without tendency to form polymer. A novel variant of staphylokinase (RGD-Sak) was designed based on the model. RGD-Sak gene was constructed by substitution mutagenesis of Lys-109 and Phe-111 with Arg-109 and Asp 111 respectively, confirmed by nucleotide sequencing. The mutant cDNA was ligated with prokaryotic expression vector pLY-4 and transformed into E. Coli JF1125. After temperature induction, over 50% expression level of RGD-Sak was achieved. RGD-Sak was isolated and purified by washing and solubilization of inclusion body, renaturation and ion exchange chromatography. The final product displayed a single band with a corresponding molecular weight of 15.5 kD in non-reducing SDS-PAGE with 95% of purity and 5×10HU/mg of specific activity. The KM、Kcat value for the activation of plasminogen with RGD-Sak was 12.40 μmol L-1 、0.81 s-1, respectively. RGD-Sak show lower tendency to polymerize after incubated with 0.9% NaCl. This study provided the basis for further reconstruction of Sak by protein engineering.
